GLP-1 & peptide half-life chart
The short answer: injectable and oral semaglutide have the longest half-life of the approved GLP-1 medications at about 1 week. Tirzepatide and dulaglutide sit near 5 days, liraglutide at about 13 hours, and immediate-release exenatide at about 2.4 hours. A dose is considered practically cleared after five half-lives, so roughly 5 weeks for semaglutide and about 25 days for tirzepatide.

How to read it
A half-life is the time it takes for half of what is in your body to be eliminated. It compounds: after one half-life about 50% remains, after two about 25%, after five about 3%. That five half-life mark is the convention behind the "practically cleared" column, and it is also roughly when levels stop climbing on a repeating schedule, which is why steady state on weekly semaglutide takes 4 to 5 weeks.
Time to peak is the other half of the picture. Semaglutide peaks 1 to 3 days after the injection, not within hours, so side effects that show up two days later are not a coincidence.
